Defragment. Then just sit back and wait, it takes between a few minutes to a few hours, depending on how long it was since you last did. I'd say yours would take a considerable amount of time.

Hmm...normally CHKDSK runs automatically when you start the computer.

Open up the Start Menu, then either select Run, and type in chkdsk.exe, or search for chkdsk.exe in the Search Programs and Files bar.
